生物絮凝剂处理高浓度细粒煤泥水的优化条件研究

摘要: 通过对白腐真菌的生物学培养特性研究,获得最佳培养条件为转速160 r/min,温度30 ℃,稻壳浸出液90 mL,葡萄糖质量浓度35 g/L,pH值3.5;同时分析了煤水中煤泥颗粒组成,发现其粒度小于0.045 mm的含量占55.93%,而大于0.25 mm的含量仅占3.75%,说明煤泥水中细粒含量较多,靠自身重力难以沉降;探讨了pH值、生物絮凝剂用量、搅拌方式和金属离子等单因素对煤泥水絮凝效果的影响,通过正交试验进一步优化了该菌所产絮凝剂对煤泥水絮凝效果的条件,结果表明:当煤泥水质量浓度27.42 g/L、助凝剂(CaCl2)质量浓度5.0 g/L、采用二段式搅拌(第一段搅拌速度60 r/min,搅拌时间180 s;第二段搅拌速度180 r/min,搅拌时间60 s)、pH值11和絮凝剂体积浓度15 mL/L时,其絮凝活性最佳,可达98.71%;进一步实验表明,絮凝活性物质绝大部分存在于细胞外,是微生物在发酵过程中菌体细胞产生的胞外分泌物。该絮凝剂具有较强的热稳定性,多种阳离子对该絮凝剂都有助凝作用,并且二价阳离子Ca2+对白腐真菌产生的絮凝剂助凝效果显著,具有良好的应用前景。

Abstract: Through to the white-rot fungus's biology raise characteristic research, has obtained the best raise condition:In rotational speed 160 r/min, the temperature 30 ℃, rice husk Lixivium 90 mL, glucose mass concentration 35 g/L, under the pH value 3.5 conditions, the fresh growing trend is best; Simultaneously has analyzed in the slime wate the granulometric composition, discovered its granularity is smaller than the 0.045 mm content to account for 55.93%, but is bigger than 0.25 mm the content only to account for 3.75%, explained in the coal spate the fine grain content are more, depends on own gravity to subside with difficulty; Discussed the pH value, the biological flocculant amount used, has stirred the way and the metallic ion and so on the single factor to the coal spate flocculation effect influence, further optimized this fungus through the orthogonal experiment to produce the flocculant to the coal spate flocculation effect condition, finally indicated:When the slime wate mass concentration 27.42 g/L, helps congeals the medicinal preparation (CaCl2) mass concentration 5.0 g/L, uses two sections of types to stir (the first section:Mixing speed 60 r/min, stirs time 180 s; Second section:When mixing speed 180 r/min, stirs time 60 s), the pH value 11 and flocculant volume concentration 15 mL/L, its flocculation activeness is best, may reach 98.71%; Further tests indicated that, the flocculation active material major part exists to the extracellular, it is the extracellular secretion which the mycelium cell produces in the fermentative process.This flocculant has a stronger thermostability, the many kinds of positive ions all are helpful to this flocculant to congeal the function, and divalent cation Ca2+ the flocculant which produces to the white-rot fungus helps congeals the effect to be remarkable, it has the good application prospect.
